Carb-X funds Fuse Diagnostics to develop innovative, instrument-free diagnostic for chlamydia and drug-resistant gonorrhea

Award to fund development and characterization of assays for Chlamydia trachomatis and Neisseria gonorrhoeae to integrate into low-cost, instrument-free diagnostic

(BOSTON: June 13, 2024) – Combating Antibiotic-Resistant Bacteria Biopharmaceutical Accelerator (CARB-X) will award up to US$1 million to Fuse Diagnostics to demonstrate proof-of-concept and feasibility ahead of the development of a rapid, instrument-free diagnostic for the detection of Chlamydia trachomatis (CT) and Neisseria gonorrhoeae (NG). Fuse is a diagnostic company that uses new, multi-analytic chemistry to develop rapid, low-cost tests.

The award will fund the demonstration of performance of their CT/NG test, Apex, on clinical samples. Apex CT/NG utilizes innovative device designs and molecular technologies with the potential to deliver accuracy that is equivalent to lab-based PCR systems in an ultra-rapid, low-cost format ideally suited to flexible global deployment, including areas with remote access to secondary healthcare in low- and middle-income countries (LMICs).

Apex CT/NG is positioned to improve current testing options, as the diagnostic’s instrument-free design aims to increase access to CT/NG diagnosis and appropriate treatment through reducing logistical complexity caused by centralized laboratory testing and the operational constraints of instrument-based platforms.

We need rapid diagnostics that allow healthcare workers to test and treat gonorrhea out of the gate, as inappropriate treatment has led to the point that only one effective antibiotic remains,” said Erin Duffy, Chief of Research and Development at CARB-X.

Gonorrhea, caused by Neisseria gonorrhoeae, is the second-most reported bacterial sexually transmitted infection, infecting approximately 82 million people globally in 2020, with the burden highest in LMICs. Patients with gonorrhea may experience serious health effects, including pelvic inflammatory disease, chronic pain, and infertility. Untreated gonorrhea can spread to the blood stream, which can be life threatening, and increase the risk of HIV infection. The true burden of gonorrhea may be greater than reported cases since patients do not always exhibit symptoms.

An estimated 1.27 million people died due to drug-resistant bacterial infections in 2019, a death toll that exceeded HIV/AIDS (864,000) and malaria (643,000) in that same year. CARB-X is building a pipeline of high-value products to prevent, diagnose and treat bacterial infections, including those that have become resistant to antibiotics. CARB-X emphasizes performance characteristics that will allow the broadest use of these products against infections driving the greatest global morbidity and mortality.

When CARB-X was founded in 2016, the early-stage antibiotic pipeline was stalled. Since then, CARB-X has supported 103 R&D projects in 13 countries, and CARB-X product developers have made tremendous progress: 18 projects have advanced into or completed clinical trials; 12 remain active in clinical development, including late-stage clinical trials; and two diagnostic products have reached the market. Additionally, at least 9 product developers with active R&D projects have already secured advanced development partnerships which can help support their clinical development after leaving the CARB-X portfolio.

In 2022, CARB-X launched new funding rounds to support R&D projects and fill critical gaps in the antibacterial pipeline. These include oral therapeutics to replace the workhorse antibiotics that are failing; vaccines for neonatal sepsis, which kills 2.5 million infants annually; and oral therapeutics, vaccines and rapid diagnostics for gonorrhea. Fuse’s diagnostic is the third diagnostic to receive a CARB-X grant as part of the 2022-2023 funding call.
