SMALL MOLECULE TARGETING PENICILLIN-BINDING PROTEINS

Innoviva Speciality Therapeutics is developing a new investigational antibiotic candidate to treat serious infections caused by multidrug‑resistant Gram‑negative bacteria, a major and growing global public health threat.

The candidate targets penicillin‑binding proteins (PBPs), essential enzymes bacteria use to construct their protective cell wall, and has been designed to overcome common resistance mechanisms. In laboratory studies, the compound has demonstrated broad-spectrum activity against a range of high‑priority Gram‑negative pathogens, including Pseudomonas aeruginosa, Klebsiella pneumoniae, Escherichia coli, Acinetobacter baumannii, and Stenotrophomonas maltophilia, as well as key resistance isolates. Infections caused by these pathogens can be life-threatening.

A recent analysis estimates that a regular release of new, potent antibiotics targeting Gram-negative bacteria could avert 11.1 million cumulative deaths caused by AMR in the coming 25 years, with the largest reductions in low-and-middle-income countries (LMICs) in regions including South Asia, Southeast Asia, East Asia and Oceania, and sub-Saharan Africa.
